Current name and lanata material

Plants of the World Online accepts Mammillaria supertexta Mart. ex Pfeiff., occurring naturally in Puebla and Oaxaca in Mexico. Mammillaria lanata (Britton & Rose) Orcutt is a synonym of the species in this database. In Lázaro-Castellanos and colleagues' 2018 study, the material bore the name M. lanata. All results described below concern this group of material, kept separate in the experiment. [2-WWW] [Lázaro-Castellanos et al. 2018, POWO, 11 October 2026. Lázaro-Castellanos et al., pp. 519–521]

Seedling development and shoot formation

Culture began with seeds supplied by the UNAM Botanical Garden. Material termed M. lanata reached about 10 mm tall after six months. It was kept on MS with 30 g/l sucrose and 0.8% agar, at 25 ± 1°C, with a sixteen-hour day and 55 µmol photons/m²/s. After initial multiplication, stem sections were obtained for the main cytokinin comparison. [Lázaro-Castellanos et al. 2018, p. 519–522]

Lateral sections on medium with meta-topolin 1.1 µM yielded a mean of 17.9 ± 1.0 shoots per explant. In the treatment without regulators, 5.6 ± 0.9 shoots formed. The most productive response was accompanied by callus on 75.0 ± 4.8% of explants. Shoots arose mainly from stimulated axillary meristems. [Lázaro-Castellanos et al. 2018, p. 522–525, tab. 4]

Rooting and acclimatisation

On MS without growth regulators, the authors obtained 94.6% rooted shoots after ten weeks. Plants about 10 mm tall were transferred to garden soil and pumice 1:1. Initial relative humidity beneath the cover was 80–90%, and after its gradual removal 50–60%. Survival during acclimatisation reported for the three groups exceeded 95%. These are results for previously rooted plants under the described greenhouse conditions. [Lázaro-Castellanos et al. 2018, p. 520–521, 524–525]

After twenty-four months in the greenhouse, 80% of the plants described in this group were flowering. They averaged about 41.2 mm tall and 30.7 mm in diameter. Age was counted from greenhouse transfer, following earlier stages of sowing, multiplication and rooting. [Lázaro-Castellanos et al. 2018, p. 524–525, ryc. 3]

Offspring in successive culture cycles

Over three cycles within two years, starting material and daughter shoots were compared using eight reproducible SSR markers. High similarity was obtained, with differences detected in some markers. Mean PIC was 0.15, 0.12 and 0.08 in successive cycles. Assessment covered eight DNA loci in the retained line of lanata material, now assigned to M. supertexta.
