From seed to pure culture
The material came from the Domeyko region in northern Chile. Seed surfaces were disinfected, then their tissues were crushed and transferred to medium. Purified cultures were derived from the resulting colonies. This yielded Arthrobacter sp. ATA002 and Cellulomonas sp. ATA003. Here, endophytic means originating from inside plant tissues. [Oses Pedraza et al. 2023] [Bartholomäus et al. 2024]
Culturing provides a living isolate and allows its DNA to be studied separately. However, the medium selected and growth conditions favour bacteria capable of developing in the laboratory. The two cultures obtained do not establish the complete composition of the seed microbiome. Culture-independent methods are also needed to investigate the broader community. [Oses Pedraza et al. 2023] [Bartholomäus et al. 2024]
Two circular sequences
Both papers used Nanopore long-read sequencing. Genome sequences were assembled from overlapping fragments, then assessed bioinformatically and genes predicted. The published sequences are genomes of bacteria cultured from seeds. [Oses Pedraza et al. 2023] [Bartholomäus et al. 2024]
| Isolate | Publication year | Genome length | GC content |
|---|---|---|---|
| Arthrobacter sp. ATA002 | 2023 | 3 904 590 base pairs | 65.9% |
| Cellulomonas sp. ATA003 | 2024 | 4 084 881 base pairs | 73.8% |
GC content specifies the proportion of the sequence represented by guanine and cytosine. Genome analysis provides a basis for comparing isolates with described bacteria and searching for genes potentially related to their function. The authors retained genus-level identifications, with the abbreviation sp. and strain number. [Oses Pedraza et al. 2023] [Bartholomäus et al. 2024]
Assembly completeness and biological function
The circular arrangement of the resulting sequence describes the outcome of DNA assembly. Additional programs assess completeness through sets of marker genes and check for possible contamination. Values depend on the selected markers and the studied organism's relationship to the reference database. Technical assessment remains part of each genome's description. [Oses Pedraza et al. 2023] [Bartholomäus et al. 2024]
The publications document the presence of living bacteria, the establishment of cultures and genomic characterisation. The effect of particular strains on germination, growth or drought tolerance requires an experiment comparing plants with the given isolate and an appropriate control. Such a stage would allow an identified gene or bacterial trait to be linked to a measurable effect on the cactus. [Oses Pedraza et al. 2023] [Bartholomäus et al. 2024]
The seed as the start of shared life
A bacterium present inside a seed is close to the tissues of the embryo and subsequent seedling. Studying its persistence during germination could clarify whether it participates in colonising the young plant. The available genomes allow strain identification to be designed and the strains tracked in such an experiment. [Oses Pedraza et al. 2023] [Bartholomäus et al. 2024]
Prepared from the publications read. Metrics, experimental conditions and tabular data were checked against the source material.

A description of the isolation of Arthrobacter sp. ATA002 from Maihueniopsis domeykoensis seeds and the sequencing of its genome. It gives the size of the assembled sequence, GC content, quality assessment and taxonomic position of the strain. The publication makes genomic data available for further research.

A description of Cellulomonas sp. ATA003 isolated from Maihueniopsis domeykoensis seeds. It presents cultivation of the isolate, Nanopore sequencing, assembly of the circular genome and its bioinformatic assessment. It includes taxonomic comparisons and accession numbers for the deposited sequences.
